A comparison of taxon-like schizotypal clusters of non-clinical individuals on polygenic scores for schizophrenia and related phenotypes

Introduction Schizotypy is conceptualized to be on the continuum of risk for psychosis. However, previous studies that used dimensional approach schizotypy failed confirm expected relations between schizotypal traits and polygenic scores (PRS) schizophrenia. A taxonic an alternative way looking at schizotypy, but best our knowledge it has not been explore genetic architecture this construct. Objectives The present study aimed fill gap by comparing groups with different profiles PRS schizophrenia related phenotypes. Methods To find clusters combinations traits, we conducted data mining ethnically homogeneous cohort 1377 healthy individuals completed Schizotypal Personality Questionnaire. Four emerged, termed low, positive, negative, high mixed schizotypy. Approximately equal from each cluster were selected genotyping. After quality control, schizophrenia, depression, neuroticism, educational attainment calculated 320 (mean age = 31.74, SD 12.25 years, 59% women) based summary statistics largest genome-wide association respective traits. then compared PRS. Results similar in sex composition differed neuroticism (as measured Eysenck Inventory), schizotypes having lowest education highest among groups. There no statistically significant differences any group showed depression. At a nominally level, negative positive low had non-significantly lower than subjects.
